Conventional Yeast Two-Hybrid VectorsWe offer two bait vectors and one library vector designed for conventional LexA- and Gal4-based yeast two-hybrid systems. Our bait vectors pLex-DIR and pDIR-Lex allow high-level, constitutive expression of baits fused either to the C-terminus or to the N-terminus of the bacterial LexA domain. Vectors carry a kanamycin marker to facilitate plasmid rescue and can be combined with most commercially available cDNA libraries. pLexA-DIR
Standard bait vector that allows fusion of your cDNA of interest to the 3’end of the LexA coding sequence. When expressed in a yeast reporter strain, the fusion protein will have the orientation LexA-BAIT.
pDIR-LexA
Reverse bait vector that allows fusion of your cDNA of interest to the 5’end of the LexA coding sequence. This may be of interest if you suspect that a free N-terminus is important for proper function of your bait. When expressed in a yeast reporter strain, the fusion protein will have the orientation BAIT-LexA. pGAD-HA
Library vector based on the popular pGAD series. cDNAs are expressed as fusions to the C-terminus of the GAL4 activation domain. pGAD-HA is a high copy vector, expression is driven from the strong ADH1 promoter. It is suitable for most other yeast two-hybrid systems. General Yeast Expression VectorsThe vectors listed here are general purpose expression vectors for use with Saccharomyces cerevisiae. They carry a KanMX marker for selection with G418. cDNAs are expressed either from a strong promoter on a high copy plasmid (p427-TEF) or from a weak promoter on a low copy plasmid (p417-CYC). p427-TEFHigh copy yeast expression vector carrying the aminoglycoside phosphotransferase gene for selection in yeast using G418. Inserts are expressed from the strong TEF promoter.
p417-CYCLow copy yeast expression vector carrying the aminoglycoside phosphotransferase gene for selection in yeast using G418. Inserts are expressed from the weak CYC1 promoter.
