Click-Chemistry
Click-chemistry was first introduced by K. Berry Sharpless in 2001. It describes a chemical reaction which builds substances by joining small units together. Click-reactions are fast and purposeful with high yields.
The most popular click-reaction is azide alkyne Huisgen cycloaddition with Copper (Cu) as catalyst at room temperature.
Now you can use this highly efficient technology for labeling of DNA or RNA via solid-phase synthesis or PCR. Furthermore, it is possible to introduce up to three different modifications onto one position of the same DNA strand. Of course, we offer different kinds of non-fluorescent and fluorescent labels.

SapphireAmp® Fast PCR Master Mix contains a hot start PCR enzyme, optimized buffer, dNTP mixture, gel loading dye (blue) and a density reagent in a 2X premix. PCR reactions with this mix can be completed in half the time required for conventional Taq amplifications. Amplification of human genomic DNA targets of 2 kb in length can be completed in 1 hour, and amplifications of human genomic DNA targets of up to 6 kb are possible using Sapphire-Amp™. This product is well-suited for E. coli-based colony PCR to check for inserts of up to ~5 kb, and colony checks can be completed in about 1 hour. SapphireAmp® Fast PCR Master Mix reactions can be assembled by simply adding primer and DNA template to the premix. Reactions performed with this mix can be loaded directly onto a gel for electrophoresis, and restriction digestion of PCR products is possible in SapphireAmp™ reaction buffer. SapphireAmp® Fast PCR Master Mix offers a powerful, fast, flexible and convenient alternative for both simple and complex PCR applications.

High Yield Gene Expression with the new T7 RNA Polymerase Gene Expression System!
This system combines the benefits of the tightly regulated and strong T7 RNA polymerase gene expression system and alkaline protease free Bacillus megaterium.
The T7 RNAP gene expression system is tightly regulated and efficiently inducible through the xylA operon and the strong T7 RNAP promoter resulting in stable, high yield protein production up to 8-fold increased in comparison to common xylose inducible expression (Fig. 1).
The system is based on two parallel-replicating plasmids: pT7-RNAP and pPT7.
In addition to the t7 rnap gene under control of the strong xylA promoter pT7-RNAP contains the genes of ampicillin and chloramphenicol for easy selection in E. coli (AmpR) and B. megaterium (CmR).
pPT7 is responsible for the T7 RNAP-dependent expression of the target gene. Downstream of the T7 promoter it comprises a multiple cloning site with ten unique restriction enzyme cleavage sites. Additionally, the plasmid carries two resistances to ampicillin (in E. coli) and tetracycline (in B. megaterium).

CellTrace™ Violet Cell Proliferation Kit (C34557)
The new gold standard in cell proliferation assays featuring CellTrace™ Violet
The class of fluorescent compounds known as cell tracking dyes is fundamental to our current understanding of the immune system and plays a major role in cancer, HIV, and post-transplant research. Several types of dyes fall into this category: some that hydrophobically insert into the plasma membrane, some which react with intracellular thiols, and others which covalently bind to amine groups inside cells. Amine-reactive cell tracking dyes, such as the new CellTrace™ Violet, are known to provide excellent long-term cell tracing. Initially a non-fluorescent ester of a fluorescent molecule, CellTrace™ Violet passively enters cells where it is converted to a fluorescent derivative by nonspecific esterases. The succinimidyl ester then covalently binds to amine groups in proteins, resulting in long-term dye retention.
When a CellTrace™ Violet-labeled cell divides, each daughter cell receives approximately half of the fluorescent label, the next generation receives one-fourth, and so on. Analysis of the fluorescence intensities of cells labeled and grown in vivo permits the determination of the number of generations through which a cell has progressed since the label was applied. This tool allows the researcher to look for dividing cells in biological systems.
The violet excitation and blue emission of CellTrace Violet will allow the researcher to perform cell proliferation experiments with a wide range of blue-excitable fluorophores, including GFP-expressing cells.

Absolute Detection of Apoptotic Transformation

What it is
The Violet Ratiometric Membrane Asymmetry Probe/Dead Cell Apoptosis Kit provides an easy, efficient method for the detection of apoptosis using a violet laser flow cytometer. The assay is compatible with adherent and suspension cell types and correlates with other indicators of apoptosis, including caspase activation and changes in mitochondrial membrane potential.
What it offers
- Robust assay - ratiometric probe is independent of probe concentration, cell size, and instrument variation
- Easy to use - 5-minute incubation, with no wash required
- Flexible experimental design - easily combined with other testing reagents
How it works
This assay employs a novel dye (F2N12S) that incorporates selectively into the outer leaflet of the plasma membrane. The dye exhibits dual fluorescence emission bands corresponding to 530 nm and 585 nm, producing a ratiometric response to variations in plasma membrane surface charge during early apoptosis that is detected by flow cytometry. The kit also includes SYTOX® AADvanced™ Dead Cell Stain for discrimination of dead cells from live cells. Unlike annexin V conjugates, this assay requires no special buffers or wash steps and is less affected by cell membrane damage commonly associated with the removal of adherent cells, thereby improving data quality.

DNAgard™ for DNA stabilization in tissues and cells
Biomatrica introduces the DNAgard™ a new product for DNA stabilization in tissues and cells. DNAgard™ stabilizes genomic DNA in cultured cells or animal tissues for at least 2 months in liquid storage format at room temperature. For longer term storage dry-down is required.
DNAgard™ is designed for immediate stabilization of DNA from intact cells and tissues with the convenience of room temperature shipping, processing and storage. The liquid storage reagent rapidly permeates cell membranes to stabilize and protect genomic DNA.
With its excellent properties DNAgard™ is ideal for field collection and tissue harvesting. Of course DNA protected using DNAgard™ is suitable for use in many downstream applications, including long-range PCR and Real Time PCR.

K-2500s PIP3 Mass ELISA Kit

K-2500s - PIP3 Mass ELISA
The K-2500s uses different PIP3 Detection protein and buffers than the
K-2500 PIP3 Mass Elisa Kit, resulting in 10x more sensitivity and selectivity to PIP3.
Class I Phosphoinositide 3-kinases (PI3-Kinase, PI3-K), are ubiquitously expressed lipid kinases which phosphorylate PtdIns(4,5)P2 at the 3'-hydroxyl of the inositol ring producing PtdIns(3,4,5)P3 (PIP3). PIP3 has shown to be an important lipid second messenger in multiple cell signalling pathways such as proliferation, metabolism, calcium release, and survival. Due to these interesting biological roles, PI3-K and PIP3 have become attractive targets for research and drug development.
The PIP3 Mass ELISA Kit allows the using to determine PI3-K activity by quantifying the amount of PIP3 found in cells. After following a non-radioactive lipid extraction protocol, the PIP3 samples obtained from millions of cells are detected on a competitive ELISA. The cellular PIP3 samples are mixed and incubated with a PIP3 Detector protein, then added to a PIP3-coated microplate for competitive binding. A peroxidase-linked secondary detector and colorimetric detection is used to detect the PIP3 protein binding to the plate. The colorimetric signal is inversely proportional to the amount of PIP3 obtained.

SensoLyteTM520 Calpain Assay
– Long Wavelength Kit
AnaSpec is pleased to announce the release of the industry’s first longest wavelength FRET based calpain assay, the SensoLyte® 520 Calpain Activity Assay Kit. Optimized for detecting calpain activity, this kit contains a novel internally quenched 5-FAM/QXL™ 520 FRET substrate. Cleavage of this FRET substrate generates two separate fragments. The subsequent release of 5-FAM fluorescence can be monitored at excitation/emission= 490/520 nm. Increase in fluorescence is proportional to the calpain activity. The assay can detect both calpain 1 (µ) and 2 (m) activities and is ideal for kinetic study of these enzymes. The long wavelength fluorescence of 5-FAM is less interfered by the autofluorescence of components in biological samples and test compounds.
Calpains are a family of intracellular cysteine proteases, which consists of at least 15 ubiquitous and tissue-specific isoforms.1 The best-characterized calpains are the isoforms µ- and m-calpain, which are also known as calpain 1 and calpain 2, respectively. Calpains are calcium-dependent and function by modulating the biological activities of many proteins through selective cleavage.2 Studies have demonstrated that calpains are implicated in a variety of calcium-regulated cellular processes such as signal transduction, cell proliferation, differentiation, cell cycle progression, apoptosis, and platelet activation. Deregulation of calpains activities has been implicated in various pathological phenomena such as atherosclerosis, Alzheimer’s disease, diabetes, and cancer.3, 4 Calpains represent potential therapeutic targets for drug development.5, 6

GR Safe Nucleic Acid Stain
More Sensitive but much less mutagen than Ethidium Bromide
GR Safe is a new, safe nucleic acid stain for detecting double-stranded DNA, single-stranded DNA, and RNA in agarose gel. It can be used for replacing mutagenic ethidium bromide (EB). GR Safe emits green fluorescence when bound to dsDNA and red fluorescence when bound to ssDNA or RNA. This new stain has two fluorescence excitation maxima when bound to nucleic acid, one centered at approximately 290 nm and one at approximately 490 nm. GR Safe is as sensitive as SYBR Green or Gelstar but much cheeper. You can use GR Safe just as the way you used EB.

TBTA - Click Chemistry Ligand for Cu (I) |

Copper (I) species are powerful catalysts for the formation of 1,2,3-triazoles from azides and alkynes. The general thermodynamic instability of Cu(I) however, results in easy oxidation to Cu(II) and/or disproportionation to Cu(0) and Cu(II).1 TBTA, Tris-[(1-benzyl-1H-1,2,3-triazol-4-yl)methyl]amine, also known as tris-(benzyltriazolylmethyl)amine, is a stabilizing ligand for Cu(I) developed by Tim Chan in Sharpless’ lab. TBTA protects Cu(I) from oxidation and dispropotionation, while enhancing its catalytic activity.1-2 A C3-symmetric derivative, TBTS is an extremely useful ligand for azide-alkyne cycloaddition with ~106 fold of rate acceleration. Its tetradentate binding ability is able to completely envelop the Cu(I) center, leaving no free binding sites available for potential destabilizing interaction. Reactions are typically carried out with 1-2 mol % Cu(I) and TBTA without having to exclude oxygen and water.3 For use as a tool in the drug discovery process, AnaSpec carries TBTA, the click chemistry reagent used in Cu(I)TBTA-catalyzed azide-alkyne cycloaddition. TBTA, with a MW of 530.6, is supplied in powder form at 97% purity.

This 16-page brochure focuses on several yeast one- and two-hybrid systems and a phagemid system, that are useful for high-throughput molecular screening of protein-protein and DNA-protein interactions. Further tools, such as biochemical assays, immunological and fluorescence-based methods, are available from MoBiTec as well. The yeast two-hybrid system has rapidly become one of the most widely used techniques in molecular biology. It still is the method of choice to identify protein-protein interactions from either cDNA libraries or known gene sequences in vivo.
